Well we can safely assume its not tobacco and I don't think Indio would be classified as your typical laid back stoner so maybe this:

CJ, I think you're on to something. The fact that Jimson weed is sometimes called locoweed would seem to make the stuff a perfect match for Indio. I note that one of the medicinal uses for the drug is to treat bedwetters: do you think Indio may have had trouble in this area and that's how he got hooked on the stuff?

From a film making perspective.
Always thought it was a
Reefer Madness
moment specially the swirling sound effect as Indio goes under.
A muddy unamed drug that takes hold of its user. And Few Dollars showing its 60s 'B Movie' pedigree.

Well reefer would be more of a calming effect , it would be more like Cheech & Chong "hey Nino man, what you mean rob the bank lets rob the chiken mole stand man,
Or imagine Nino knocking at a shut door " Hey Indio man, open up its me Nino I got the stuff". Indio on the other side "Nino's not here", etc., etc.,

CJ is right about the effects of reefer (how does he know this so well?). Also his suggestion that Jimson weed is the likely culprit seems a good one. Consider this from an entry at wikipedia:
"The effects of Datura [Jimson weed] have been described as a living dream: consciousness falls in and out, people who don't exist or are miles away are conversed with etc. The effects can last for days. Tropane alkaloids [one of the active ingredients] are some of the few substances which cause true hallucinations which cannot be distinguished from reality. This is unlike psylocybin or LSD, which only cause visual distortions.
"The doses that cause noticeable effects, and the doses that can kill are very close with datura. This makes overdosing on Datura stramonium very easy. This can be fatal, and it can cause fevers in the 105-110 range which is a range that can kill brain cells, and lead to brain damage."
Hmmm, brain damage? Could explain a lot about Indio, don't you think?

Maybe, but Jimson weed can account for all of Indio's symptoms, not just his behavior. For example, the hallucinations Indio experiences (the flashbacks popularly thought to be merely memories) could be the equivalent of seeing "people who don't exist or are miles [or years] away," as cited above.
