For me, it was the Ecstacy of Gold sequence in GBU at first viewing (June 1968), after over 2.25 hours into the film.

For a Few Dollars More....the Mortimer Guy Calloway sequence  Afro

Why, cj?

That was the first Leone film I saw and from when Mortimer arrives at the hotel in Tucumcari to when he shoots down Calloway in the street was like nothing I've ever seen in a Western.

For me, Once Upon a Time In the West is one long defining moment.
Not nearly long enough, but defining.

In For a Few Dollars More. This was a "B" movie which meant it played second at the drive in I was at. I had a date so I wasn't much interested in the movie
if you know what I mean. Then came the shootout with Eastwood and the 3 guys standing in the door of the saloon. I forgot the girl and watched the rest
of the movie. Been an Eastwood spaghetti western fan every since.

All of the showdowns in each of the dollars movies.
Ecstasy of gold sequence.
El Indio killing that man's family in the church then challenging him to a duel.
Juan coming back to the grotto to see his family murdered, and John coming in like a badass and saving him from the firing squad.
The showdown of OUATITW.
The final conversation between Noodles and Max in OUATIA.
