I also like the scene in the monastery where Tuco is asking Blondie, "Tell me?  Tell me?, then Blondie asks Tuco to "Come closer".  Tuco thinks Blondie is going to tell him where the gold is (but we all know Blondie's much smarter than that; Tuco can't fool him), getting all excited, then gets coffee thrown in his face - VERY funny scene indeed - cracks me up all the time -  ;D O0

After Tuco is telling Blondie about his brother and the bowl of soup and Blondie says after a good meal there nothing like a good cigar................great sense of pathos

That's one of the greatest moments of the film, I agree. O0

The majority of Tuco's lines mostly. here's a few favorites.

"When you gotta shoot, shoot, don't talk."
"I like big fat men like you, when they fall they make more noise and sometimes they never get up."
"Roll it up, roll it up! I'll give you a good idea where you can put it!"

I also like some of the Union captains lines, he was great. "Whoever has the most liquor to get the soldiers drunk and send them to be slaughtered... he's the winner. "

Angel Eyes' line when Tuco is receiving his beating really had me going though. "How's your digestion now?" I'm a big Tuco fan and all but that was priceless.

"Yeah, but you don`t look like the one who will collect it. Couple of steps back." - Blondie

"You know whose son you are? You don`t , I do, everybody does......" - Tuco

"I am glad they got him. A man guilty of all those crimes." - Lady in the stagecoach

"People with ropes around their necks don`t always hang." - Angel Eyes

"One name is as good as any other. Not wise to use your own name, like you, I bet they don`t call you Angel Eyes...Sergeant Angel eyes." - Tuco

Is it wrong to speak the way I do to volunteers? I've done a lot worse.

"When you've got to shoot, shoot...don't talk!" --Tuco.  It's been mentioned already by other people, but it's always been a favorite.  Hell, I could write down the whole script before running out of favorite lines.

Speaking of that, do deleted scene lines count?  I like the one where Blondie is 'camping' with Angel Eyes and the men come out of the woodwork, Blondie counts them, and mentions something about that perfect number in relation to the bullets in his gun.   ;D

Must re-watch. Not like I haven't seen it numerous times...  ;D But the quotes always leave me wanting more.

Yes there are many quotes, but I like when Angel Eyes asks Tuco if he likes music with his food and Tuco says Music yes, is very good for the digestion.  Tuco does a great job in that scene, just watch his eyes move when he answers Angel Eyes, it's great. I also like when Blondie and Tuco are in the carriage together and Tuco yells Hurray down with General Grant Hurray for the Confederacy Hurray for General ?  And Blondie tells him Lee.

Tuco: "When you have to shoot.. shoot, don't talk".
Blondie: "You see in this world there's two kinds of people, my friend. Those with loaded guns, and those who dig. You dig."
Tuco: "You hear, that? They say even when Judas hung himself it thundered."
Tuco to priests: "Take care of him he's like a brother to me." or  "Father, you don't know how much this boy's life means to me."

Think of how you'd be if you'd been on your own.
